What Are Natural Killer Cells (NK Cells)?
They are part of your immune cells that are your body’s frontline defense. This army consists of: Natural Killer, T cells, macrophages, and dendritic cells. You are born with them, and their primary function is to kill infected and tumor cells, hence the name.
In recent years, science has been uncovering the powerful, complementary role that Natural Killer (NK) cells play in breast cancer healing. There are also a few Natural Killer Cell-specific therapies, as well as many more in development, that may provide remarkable results.

- NK cells don’t require an antigen to recognize a target. They can act without prior sensitization, making them faster responders.

- There are subtypes of NK Cells. For instance, CD56^bright NK cells (more regulatory / cytokine producers) and CD56^dim NK cells (more cytotoxic, found in peripheral blood).
- NK cells don’t rely on T-cell receptors (unlike T cells). They can sometimes be used in “allogeneic” or donor settings (less risk of rejection) in immunotherapy design.

How NK Cells Detect And Kill Cancerous Cells
NK cells use a delicate balance of “on” and “off” signals to decide whether a cell is dangerous. If the inhibitory signals dominate, they stand down. Even better, if activating signals dominate (i.e., the cell is stressed, mutated, or showing unusual markers), NK cells can deliver a lethal hit.
Mechanisms of NK cell cytotoxicity include:
- Perforin + Granzymes: NK cells release perforin, which creates pores in the target cell membrane. Through those pores, granzymes (proteases) enter and trigger apoptosis (programmed cell death).
- Death receptor pathways: NK cells can engage death receptors (like Fas) on target cells, triggering internal death cascades.
- Cytokine production: NK cells secrete IFN-γ and other cytokines that help recruit and activate other immune cells (macrophages, T cells).
- Antibody-dependent cellular cytotoxicity (ADCC): via CD16 receptor, NK cells may bind antibodies coating a tumor cell and kill it.
Because they don’t require that a tumor cell present a specific antigen (unlike T cells), NK cells are uniquely positioned to catch cells that have evolved to evade the more specialized immune surveillance.
Benefits of Natural Killer Cells for Breast Cancer Healing

Groundbreaking Study: NK Cells Can Stop Metastasis
In 2020, a first-of-its-kind, groundbreaking study [8] found that NK Cells can be reprogrammed to stop metastasis. As we mentioned above, science has only recently begun to tap into the power of NK Cells. There is still much to research, but we are off to a promising start. In any case, this study employed a novel 3-D cell culture method to observe the interactions between NK cells and invasive breast cancer cells in real-time. The researchers observed that NK cells activate an enzyme in K14+ cells, causing them to undergo cell death. Meaning that when used strategically, NK Cells limit early metastasis.
